A new study published in the journal Oncoscience has identified a promising approach to treating glioblastoma, one of the most aggressive forms of brain cancer.
Researchers found that NO-Cbl, a modified form of vitamin B12 designed to release nitric oxide, can cross the blood-brain barrier and accumulate selectively in tumour tissue.
The research team, led by Joseph A Bauer of Nitric Oxide Services, LLC, and the Cleveland Clinic Foundation’s Taussig Cancer Centre, investigated whether NO-Cbl could improve drug delivery to glioblastoma, where the blood-brain barrier often limits the effectiveness of treatment.
Scientists tested the compound using cancer cell lines, rat models and combination therapies.
While NO-Cbl alone showed moderate activity against glioblastoma cells, it demonstrated broad anti-tumour effects across several cancer types.
Animal studies produced one of the most significant findings.
Researchers found that NO-Cbl crossed the blood-brain barrier after systemic administration and remained concentrated in glioblastoma tissue for at least 24 hours, while healthy tissues cleared the compound much more quickly.
The team also combined NO-Cbl with existing glioblastoma treatments, including temozolomide and TRAIL.
Laboratory experiments on human glioblastoma cells showed that the combination suppressed tumour growth more effectively than either treatment alone.
According to the researchers, NO-Cbl may help overcome treatment resistance by triggering cancer cell death while making tumour cells more responsive to existing therapies.
Although the findings remain preliminary, the study suggests that NO-Cbl could become a promising addition to future glioblastoma treatment strategies.
Further research will focus on confirming the results and determining safe and effective dosing for clinical use.
