Routine childhood immunisations do not increase the risk of epilepsy in young children, according to a large study led by the US Centres for Disease Control and Prevention (CDC).
The findings provide fresh reassurance for parents amid ongoing concerns about vaccine safety.
Published in The Journal of Paediatrics, the study also found no evidence that aluminium-based adjuvants used in vaccines raise the likelihood of developing epilepsy during early childhood.
Researchers analysed health data from 2,089 children diagnosed with epilepsy between the ages of one and four years. These cases were compared with 20,139 children without epilepsy, matched by age, sex and healthcare provider.
More than half of the children in the study were boys, and nearly 70 per cent fell between the ages of one year and 23 months. The analysis found no higher epilepsy risk among children who followed the recommended vaccination schedule.
To assess vaccine exposure, the team reviewed routine immunisation schedules and calculated cumulative aluminium exposure from vaccine adjuvants, measured in milligrams.
Aluminium salts, commonly used to enhance immune response, have previously been the subject of safety debates.
However, the researchers reported that staying up to date with vaccinations and cumulative aluminium exposure did not increase the risk of epilepsy. Adjusted odds ratios for both factors remained below levels of concern.
The study found that children with known risk factors, such as premature birth, a family history of epilepsy, or underlying neurological or medical conditions, had significantly higher odds of developing epilepsy, independent of vaccination status.
A subgroup analysis suggested that infants aged one to two months who received vaccines containing a specific aluminium adjuvant combination showed higher odds of an epilepsy diagnosis. However, these findings did not reach statistical significance.
Researchers concluded that the findings add to existing evidence supporting the safety of childhood vaccination programmes, particularly at a time when immunisation coverage has declined in some communities.
They noted that the results could help healthcare providers address parental concerns and reinforce confidence in routine childhood vaccines.
