UQ Researchers Study Lung Cancer Cell ‘Neighbourhoods’ To Explain Immunotherapy Failure

Researchers from the University of Queensland (UQ) Frazer Institute have investigated the intricate ‘neighbourhoods’ of non-small cell lung carcinoma (NSCLC) cells to understand why some patients fail to respond to immunotherapy.

By examining cell interactions and glucose metabolism at cellular resolution, the team identified metabolic patterns linked to treatment response and resistance.

Employing advanced machine-learning algorithms and computational models, they analysed how cancer cells metabolise glucose, a nutrient crucial to tumour growth.

Associate Professor Arutha Kulasinghe explained that their study delved into the ‘personal lives’ of cells within a tumour, revealing that specific metabolic microenvironments significantly influence whether immunotherapy succeeds or fails.

Lead author James Monkman noted that regions where cancer cells took up more glucose showed poorer outcomes, explaining that glucose processing differs across tumour regions and directly influences treatment response.

The study underscores the importance of identifying patients most likely to benefit from immunotherapy, as well as those who may require alternative or combination treatments.

Published in Nature Communications, the research provides a foundation for developing metabolic inhibitors and advancing precision medicine tailored to each patient’s tumour profile.

The team aims to extend this approach to other cancers, offering the potential for more effective and personalised therapies.

By targeting cancer cell metabolism, clinicians could moreover improve patient outcomes, reduce treatment costs, and provide therapies customised to the unique composition of each tumour, marking a significant step forward in precision oncology and enhancing the effectiveness of immunotherapy for patients with resistant cancers.
