Researchers developed a promising HIV vaccine that trains the immune system to produce powerful antibodies against multiple virus strains.
The study, published in ‘Nature’, was led by scientists from the La Jolla Institute for Immunology (LJI) and Scripps Research.
Unlike traditional vaccines, this approach activates rare immune cells early and strengthens them through carefully timed booster doses.
Over time, these cells produce broadly neutralising antibodies that target stable parts of HIV, helping protect against many rapidly changing virus strains.
To test the strategy, researchers designed special protein-based vaccine components that closely resemble key parts of HIV.
Researchers further gave rhesus macaques an initial vaccine dose followed by several booster shots.
Each booster helped the immune cells mature step by step, strengthening their ability to produce broader protection against different HIV strains.
Patrick Madden said the vaccine gradually guides B cells to produce broadly neutralising antibodies that can fight multiple HIV strains.
Over half the animals developed memory B cells, while 44% produced broadly neutralising antibodies, showing strong early vaccine success.
One animal-generated antibody level that researchers believe could protect against a wide range of HIV strains.
The vaccine’s first dose has already been tested in the HVTN 144 clinical trial and is currently being evaluated in the ongoing Phase I IAVI G004 trial.
Researchers, including LJI professor Shane Crotty, believe the approach may work even better in humans because of differences in the human immune system.
Scientists will now focus on improving the booster sequence and increasing response rates.
The findings provide strong evidence that this vaccine strategy could bring the world much closer to developing a safe and effective HIV vaccine.
